Creatinekinase(CK1)playsanimportantroletransferringphosphategroupsfromphosphocreatinetoADPinthecytosoloftissueshavinghigh,fluctuatingenergydemandslikeskeletalmuscle,heart,andbrain. MitochondrialCK1,CK1Mt,hastwoisoforms,sarcomericCK1MtandubiquitousCK1Mt. CK1MtislocalizedbetweentheMIM(mitochondrialinnermembrane)andMOM(mitochondrialoutermembrane)andboundtothecardiolipin-richinnerleaflet(Mulleretal.,1985)andalongthecristaemembranes(Wegmannetal.,1991). CK1Mtexistsintwoforms;ahomodimerandanoctamerconsistingoffourhomodimers. TheCK1MtoctamerformspermanentcontactsitesandmaintainsacomplexstructureincludingporinandANT(adeninenucleotidetranslocase)withinMIMandMOM(Speeretal,2005).Therehavebeenseveralphospho-serine,threonine,andtyrosinesitesidentifiedwithinCK1Mt,theroleofeachonehasyettobedetermined.
