Alpha-internexinisaClassIVintermediatefilamentoriginallydiscoveredasitco-purifieswithotherneurofilamentsubunits(1).Alpha-internexinisrelatedtobutdistinctfromthebetterknownneurofilamenttripletproteins,NF-L,NF-MandNF-H,havingsimilarproteinsequencemotifsandasimilarintronorganization.Itisexpressedonlyinneuronsandinlargeamountsearlyinneuronaldevelopment,butisdown-regulatedinmanyneuronsasdevelopmentproceeds.Manyclassesofmatureneuronscontainalpha-internexininadditiontoNF-L,NF-MandNF-H.Insomematureneuronsalphainternexinistheonlyneurofilamentsubunitexpressed.Antibodiestoalpha-internexinarethereforeuniqueprobestostudyandclassifyneuronaltypesandfollowtheirprocessesinsectionsandintissueculture.Inaddition,recentstudiesshowamarkedup-regulationofalpha-internexinduringneuronalregeneration(2).Theuseofantibodiestothisproteininthestudyofbraintumorshasnotbeenexaminedtodate,butislikelytobeofinterest.RecentlyCairnsetal.usedthisantibodytoshowthatalphainternexinisanabundantcomponentoftheinclusionsofneurofilamentinclusionbodydisease(NFID),aserioushumanneurodegenerativedisorder(3,4).Theantibodywasalsousedtoconfirmthepresenceofcirculatingauto-antibodiestoalpha-internexinintheseraofsomepatientswithendocrineautoimmunity,aswellasinsomenormalindividuals(5).
