ATRIP,ATRinteractingprotein,bindstoATRtoregulateATRexpression,andisanessentialcomponentoftheDNAdamagecheckpointpathway(Cortezetal,2001).ATRisrecruitedtoDNAlesionsinpartthroughitsassociationwithATRIP,whichinturninteractswiththesingle-strandedDNAbindingproteinRPA(Balletal,2007).DNAreplicationforksmaystallasaresultofDNAdamagecausingphosphorylationofseveralproteins,includingBRCA1whencolocalizingwithATR/ATRIPcomplexandRPA(Venereetal,2007).TheDNAreplicationforkstallcoincideswithBRCA1directlyphosphorylatingATRIPatSer239(Venereetal,2007).
