CtIP,C-terminalbindingprotein-interactingprotein,isaDNAendonucleaseactivatedbydoublestrandedbreaks(DSBs). DSBrepairscanbeperformedbyeitheroneoftwomechanisms;non-homologousendjoining(NHEJ)orhomologousrecombination(HR). NHEJisthepredominantDSBrepairpathwaythroughouttheentirecellcycle,mostimportantlyintheG1phase(Rothkammetal,2003);whileHRisimportantforrepairingDSBsinSandG2phases(Beucheretal,2009).CtIPcontrolsDSBresection;aneventthatonlyoccursinHRduringG2-phase.PhosphorylationofThr847dictatestheresectionefficiency(Huertasetal,2008).FurThermore,ithasbeenfoundthatDSBsundergoresectionandrepairinG1-phasecellsviaaprocessrequiringPlk3phosphorylationofCtIPatSer327andThr847(Bartonetal,2014). SeveraladditionalphosphorylationsiteswithinCtIPhavebeenidentified,buttheirsignificanceintherepairofDNAhaveyettobedetermined.
