TheDrosha-DGCR8microprocessorcomplexisrequiredformicroRNA(miRNA)biogenesis.DGCR8(DiGeorgeSyndromeCriticalRegion8)recognizestheRNAsubstrate,whereasDroshafunctionsastheendonuclease.DGCR8,whichcontainstwodouble-strandedRNA(dsRNA)-bindingdomains,interactswiththepri-miRNAandfunctionsasthemolecularanchorthatmeasuresthedistancefromtheds-RNA-ssRNAjunctionanddirectsDroshacleavage11bpaway(HanJetal,2006).TheefficiencyofDroshacleavageincreasesinthepresenceofhemeandpromotestheformationofhighlyorderedDGCR8structuresuponbindingtoRNA(Falleretal,2010).
