Fanconianemia,FA,isararedisorderwherecellscannotprevent,repair,ortolerateDNAdamage,leADIngtocancer,progressivebonemarrowfailureanddevelopmentalabnormalities(Ishiaietal.,2008). 16geneshavebeenimplicatedinFA,andtheirproductsconstituteacommonFApathway(WaldenandDeans2014). FANCI(FanconianemiacomplementationgroupI),isoneoftwosubstratesformonoubiquitnationbytheFANCL-containingcorecomplex,andiscrucialforDNArepairviaFAPandintrastrandcross-links(WaldenandDeans,2014).Phosphorylationofser556andser559arehighlyconservedandhavebeendeterminedtobeATM/ATRkinasesubstratesfortheFA-DNApathway(Smogorzewskaetal.,2007). FurThermore,thephosphorylationofser556andser559havebeenshowntoplaykeyrolesinFANCIphysicallyassociatingwithFANCD2,stABIlizingtheID2complex,andsupportingitsDNAbindingandsubsequentD2-ubiquitination(WaldenandDeans,2014). ResearchcontinuestodetermineifonesiteplaysmoreofaroleintheDNArepair/damagecheckpointoftheFApathwayandstabilizationoftheID2complex.
