Gamma-aminobutyricacid(GABA)istheprimaryinhibitoryneurotransmitterinthecentralnervoussystem,causingahyperpolarizationofthemembranethroughtheopeningofaCl−channelassociatedwiththeGABAAreceptor(GABAA-R)subtype.GABAA-Rsareimportanttherapeutictargetsforarangeofsedative,anxiolytic,andhypnoticagentsandareimplicatedinseveraldiseasesincludingepilepsy,anxiety,depression,andsubstanceabuse.TheGABAA-Risamultimericsubunitcomplex.Todatesixαs,fourβsandfourγs,plusalternativesplicingvariantsofsomeofthesesubunits,havebeenidentified(OlsenandTobin,1990;Whitingetal.,1999;Ogrisetal.,2004).InjectioninoocytesormammaliancelllinesofcRNAcodingforα-andβ-subunitsresultsintheexpressionoffunctionalGABAA-RssensitivetoGABA.However,coexpressionofaγ-subunitisrequiredforbenzodiazepinemodulation.Thevariouseffectsofthebenzodiazepinesinbrainmayalsobemediatedviadifferentα-subunitsofthereceptor(McKernanetal.,2000;MehtaandTicku,1998;Ogrisetal.,2004;Pöltletal.,2003).
