Theantibodyispreparedfromrabbitserumbyaffinitypurificationviasequentialchromatographyonphospho-anddephospho-peptideaffinitycolumns.
BIOLOGicalSignificance:Collapse
Theenzyme5-lipoxygenase(5-LO)playsakeyroleinregulatingtheproductionofleukotrienes(LTs)(Funk,2001).OverproductionofLTscontributestoseveraldiseases,mostnotablychronicinflammatorydiseases,includingasthma(Drazenetal.,1994),fibrosis(Wilbornetal.,1996)andatherosclerosis(Dwyeretal.,2004).Recentworkhasdemonstratedthattheactivityof5-LOisregulatedbyPKAphosphorylationofserine-523in5-LO(Luoetal.,2004).Undernormalconditions,thisphosphorylationmaybeimportantinlimitinginflammation.AbnormalsignalingthroughcAMPandPKA,then,couldcontributetoavarietyofdiseases,includingthosecharacterizedbychronicinflammation.Thephospho-specificantibodytoSer523on5-LOisthuslikelytoprovideavaluabletoolforstudiesoftheroleof5-LOregulationindiseasessuchasasthma,fibrosisandatherosclerosis.
