BIOLOGicalSignificance:Collapse
Tryptophanhydroxylase(TPH)catalyzesthe5-hydroxylationoftryptophan,whichisthefirststepinthebiosynthesisofindoleamines(SEROtoninandmelatonin)(Martinezetal.,2001).Inmammals,serotoninbiosynthesisoccurspredominantlyinneuronswhichoriginateintheRaphenucleiofthebrain,andmelatoninsynthesistakesplacewithinthepinealgland.AlthoughTPHcatalyzesthesamereactionwithintheRaphenucleiandthepinealgland,TPHactivityisrate-limitingforserotoninbutnotmelatoninbiosynthesis.Serotoninfunctionsmainlyasaneurotransmitter,whereasmelatoninistheprincipalhormonesecretedbythepinealgland.TheactivityofTPHisenhancedbyphosphorylationbycAMP-dependentproteinkinase(PKA)andCa2+/calmodulinkinaseII(CaMKII)(Jiangetal.,2000;Johansenetal.,1996)BothPKAandCaMKIIphosphorylateSer58whichlieswithintheregulatorydomainofTPH(Kuhnetal.,1997).
