Theandrogenreceptor(AR)isaDNA-bindingtranscriptionfactorthatregulatesgenescriticalforthedevelopmentandmaintenanceofthemalesexualphenotype.Defectsinandrogenreceptorhavebeenshowntoplayaroleinprostatecancer,andinhibitionofARactivitythroughmodulationofsignaltransductionpathwaysmaydelayprostatecancerprogression(HeinleinandChang2004).Multiplephosphorylationsiteshavebeenidentifiedontheandrogenreceptorthataffectcross-talkbetweengrowthfactorsignalingandandrogeninprostatedevelopmentandcancer(Gioelietal.,2002).Oneofthesesites,atSer94,appearsconstitutivelyphosphorylatedandexhibitsnoresponsetotreatmentswithstimulatinghormone(Gioelietal.,2002).ThesiteatSer94isuniqueamongtheARphosphorylationsitesinthatitdoesnotachieveamaximallevelofphosphorylationbetweentranslationandtheinitialroundofnuclearimport,havingastrongbiasforandrogen-independentphosphorylationinthecytoplasm(Kesleretal.,2007).
