Thereisaccumulatingevidencetosuggestthatprogesteroneplaysanessentialroleintheregulationofgrowthanddifferentiationofmammaryglandsandthusmayplayakeyroleinbreastcancer(Edwards,2005).Thebiologicalresponsetoprogesteroneismediatedbytwodistinctformsofthehumanprogesteronereceptor(PR-AandPR-Bforms).Inmostcellcontexts,theBformfunctionsasatranscriptionalactivator,whereastheAformfunctionsasatranscriptionalinhibitorofsteroidhormones(Attiaetal.,2000;Linetal.,2003).RecentlyithasbeendemonstratedthatthereisdifferentialhormonedependentregulationofthephosphorylationoftheAandBformsofthereceptor(Clemmetal.,2000).TreatmentofT47DbreastcancercellswithprogestinagoNISTincreasesthephosphorylationofSer190andSer294withdifferentkinetics.Thesephosphorylationeventsmaydifferentiallyaffectthetranscriptionalactivityofthereceptor.
