Rhodopsinisaphotoreceptorproteinfoundinretinalrods.Itisacomplexformedbythebindingofretinaldehyde,theoxidizedformofretinol,totheproteinopsinandundergoesaseriesofcomplexreactionsinresponsetovisIBLelightresultinginthetransmissionofnerveimpulsestothebrain.Mutationoftherhodopsingeneisamajorcontributortovariousretinopathiessuchasretinitispigmentosa.Thedisease-causingproteingenerallyaggregateswithubiquitinininclusionbodies,disruptstheintermediatefilamentnetworkandimpairstheABIlityofthecelltodegradenon-functioningproteinswhichleadstophotoreceptorapoptosis(Bersonetal.,1991).OthermutationsonrhodopsinleadtoX-linkedcongenitalstationarynightblindness,mainlyduetoconstitutiveactivation,whenthemutationsoccuraroundthechromophorebindingpocketofrhodopsin(Dryjaetal.,1993).Severalotherpathologicalstatesrelatingtorhodopsinhavebeendiscoveredincludingpoorpost-Golgitrafficking,dysregulativeactivation,rodoutersegmentinstabilityandarrestinbinding.
