Nop1pwasoriginallyidentifiedasanucleolarproteinofbakersyeast,Saccharomycescerevisiae.TheNop1pproteinis327aminoacidsinsize(34.5kDa),isessentialforyeastviABIlity,andislocalizedinthenucleoli(1).ThesystematicnameforS.cerevisiaeNop1isYDL014W,anditisnowknowntobepartofthesmallsubunitprocessomecomplex,involvedintheprocessingofpre-18SribosomalRNA.Nop1pistheyeasthomologueofaproteinfoundinalleukaryotesandarchaeagenerallycalledfibrillarin(2).Fibrillarin/Nop1pisextraordinarilyconserved,sothattheyeastandhumanproteinsare67%identical,andthehumanproteincanfunctionallyreplacetheyeastprotein.Patientswiththeautoimmunediseasesclerodermaoftenhavestrongcirculatingautoantibodiestoa~34kDaproteinwhichwassubsequentlyfoundtobefibrillarin.Recentstudiesshowthatknock-outofthefibrillaringeneinmiceresultsinembryoniclethality,althoughmicewithonlyonefunctionalfibrillarin/Nop1pgenewereviable(3).ThisantibodyisbecomingwidelyusedasaconvenientMarkerfornucleoliinawidevarietyofspecies(e.g.4-6).
