GlialFibrillaryAcidicProtein(GFAP)wasdiscoveredbyAmicoBignamiandco-workersasamajorfibrousproteinofmultiplesclerosisplaques(1).Itwassubsequentlyfoundtobeamemberofthe10nmorintermediatefilament(IF)family,specificallytheIFfamilyClassIII,whichalsoincludesperipherin,desminandvimentin.GFAPisstronglyandspecificallyexpressedinastrocytesandcertainotherastrogliaintheCNS,insatellitecells,peripheralganglia,andinnon-myelinatingSchwanncellsinperipheralnerves.InmanydamageanddiseasestatesGFAPexpressionisheavilyupregulatedinastrocytes.Inaddition,neuralstemcellsfrequentlystronglyexpressGFAP.PointmutationsintheproteincodingregionoftheGFAPgeneleadtoAlexanderdiseasewhichischaracterizedbythepresenceofabnormalastrocytescontainingGFAPproteinaggregatesknownasRosenthalfibers(2).
