Microtubulesare25nmdiameterproteinrodsfoundinmostkindsofeukaryoticcells.Microtubulesareassociatedwithafamilyofproteinscalledmicrotubuleassociatedproteins(MAPs),whichincludestheprotein(tau)andagroupofproteinsreferredtoasMAP1,MAP2,MAP3,MAP4andMAP5(Kindler&Gardner1994).MAP2ismadeupoftwo~280kDabandsreferredtoasMAP2aandMAP2b.Athirdlowermolecularweightform,MAP2CandMAP2D,correspondstoapairofproteinbandsrunningat~70kDaonSDS-PAGEgels.AlltheseMAP2formsarederivedfromasinglegenebyalternatetranscription,andallshareaC-terminalsequencewhichincludeseitherthreeorfourmicrotubulebindingpeptidesequences,whichareverysimilartothosefoundintherelatedmicrotubulebindingprotein(tau).MAP2isoformsareexpressedonlyinneuronalcellsandspecificallyintheperikaryaanddendritesofthesecells.MAP2CandMAP2DareexpressedearlierindevelopmentthantheMAP2aandMAP2bisoforms,sothatthisantibodyisamoreusefulMarkerofneuronaldevelopment.MAP2hasbeenrecentlyshowntobethespecificreceptorfortheneurosteroidpregnenolone(FontaineLenoreV.etal.,2006).
