BIOLOGicalSignificance:Collapse
MyelinBasicProtein(MBP)isoneofthemajorproteinsofthemyelinsheathsurroundingaxonsinthenervoussystem.Sinceitisofrelativelylowmolecularweightandhighabundancetheproteinsequencewasdeterminedfrompurifiedproteinover30yearsago(Eylaretal.,1971).Theproteinismadebyoligodendrocytesinthecentralnervoussystem,soantibodiestoMBParegoodMarkersofthiscelltype.However,transcriptsfromthesamegenearealsoexpressedincertainhematopoeticlineagecells(Martyetal.,2002).Inthecentralnervoussystemtherearetheredifferentformsoftheproteinmadebyalternatetranscriptionfromasinglegene,whichhavemolecularweightsof21.5,18.5,and17.2kDa.Sincethetwolowermolecularweightformsareverycloseinmolecularsize,MBPantibodiestypicallyshowtwobandsonWesternblots,oneatabout22kDaandanotheratabout18kDa.
