NestinisamemberoftheclassIVintermediatefilamentproteinfamilywhichisexpressedinneuronalstemcells(Lendahletal.,1990).Nestinwasoriginallyidentifiedasaresultoftheproductionofaseriesofmonoclonalantibodiesdirectedagainstepitopesexpressedonformalinfixedembryoday15ratspinalcord(Hockfieldetal.,1985)).Oneoftheseantibodies,calledRat401,stainedfibrousprofilesinthedevelopingnervoussystem,butnotinthematurenervoussystem.ByscreeningbacteriophageexpressionlibrarieswithRat401,Lendahletal.wereabletoisolateaCDNAencodingtheproteintowhichRat401antibodybound.Theproteinprovedtobeanunusualmemberoftheintermediatefilamentfamily,containinganalpha-helicalregionhomologoustothatfoundinkeratinandneurofilamentsubunits.
