BIOLOGicalSignificance:Collapse
TDP43(TarDNABinding43,TARDBP)wasoriginallyidentifiedasaproteinwhichbindstothe“transactivationresponse”(TAR)sequencefoundinthelongterminalrepeatoftheHIV-1virusgenome(Ouetal.,1995).UVcross-linkingofHeLacellextractrevealeda43kDaproteinwhichwasclonedandsequencedandshowntocontaintwocopiesofthe~90aminoacidRRMdomain.RRMisanacronymforRNARecognitionMotif,andthisdomainisfoundinmanyproteinswhichbindsinglestrandedRNAandsomewhichbindsinglestrandedDNA.Northernblotsshowedthattheproteinisubiquitousintissueexpression.MuchinteresthasbeenfocusedonTDP43recentlyduetoitsassociationwiththeinclusionsseeninfrontotemporallobardegenerationandAmyotrophicLateralSclerosis(Neumannetal.,2006).Theproteinispresentintheseinclusionsinpartiallydegraded,hyperphosphorylatedandubiquitinatedforms.
