UbiquitinC-terminalhydrolase1(UCHL1)isalsoknownasubiquitincarboxylesteraseL1,ubiquitinthiolesterase,neuron-specificproteinPGP9.5andPark5.Itwasoriginallyidentifiedasamajorcomponentoftheneuronalcytoplasmfrom2-dimensionalgelanalysisofbraintissues,andwasgiventhenamePGP9.5(1).ItwaslaterfoundthatubiquitinC-terminalhydrolaseenzymeactivitywasassociatedwiththePGP9.5protein(2).TheubiquitinC-terminalhydrolasescleaveubiquitinfromothermolecules.Regulationoftheubiquitinpathwayisveryimportantandmanydiseasestatesareassociatedwithdefectsinthispathway.GeneticknockoutofUCHL1inmiceresultsinamotorneurondegenerationsimilartothespontaneousgracileaxonaldystrophy(gad)mutantmice(3).PointmutationsintheUCHL1geneareassociatedwithsomeformsofhumanParkinson’sdisease(4).SinceUCHL1isheavilyexpressedinneurons,itisreleasedinlargeamountsfollowinginjuryordegeneration,sothedetectionofUCHL1inCSFandotherbodilyfluidscanbeusedasabioMarker.
