BIOLOGicalSignificance:Collapse
Gamma-aminobutyricacid(GABA)istheprimaryinhibitoryneurotransmitterinthecentralnervoussystem.TherearetwomajorclassesofGABAreceptors:theGABAA andtheGABAB subtypeofreceptors.GABAA-Rsareimportanttherapeutictargetsforarangeofsedative,anxiolytic,andhypnoticagentsandareimplicatedinseveraldiseasesincludingepilepsy,anxiety,depression,andsubstanceabuse.TheGABAA-Risamultimericsubunitcomplex.Todatesixαs,fourβsandfourγs,plusalternativesplicingvariantsofsomeofthesesubunits,havebeenidentified(OlsenandTobin,1990;Whitingetal.,1999;Ogrisetal.,2004).InjectioninoocytesormammaliancelllinesofcRNAcodingforα-andβ-subunitsresultsintheexpressionoffunctionalGABAA-RssensitivetoGABA.However,coexpressionofaγ-subunitisrequiredforbenzodiazepinemodulation.Phosphorylationofserine408andserine409withintheβ3subunithavebeenshowntobecriticalforthefunctionalmodulationofβ3containingrecombinantreceptors(Brandonetal.,2000).
