Gamma-aminobutyricacid(GABA)istheprimaryinhibitoryneurotransmitterinthecentralnervoussystem.TherearetwomajorclassesofGABAreceptors:theGABAAandtheGABAABsubtypeofreceptors.GABAA-Rsareimportanttherapeutictargetsforarangeofsedative,anxiolytic,andhypnoticagentsandareimplicatedinseveraldiseasesincludingepilepsy,anxiety,depression,andsubstanceabuse.TheGABAA-Risamultimericsubunitcomplex.Todatesixαs,fourβsandfourγs,plusalternativesplicingvariantsofsomeofthesesubunits,havebeenidentified(OlsenandTobin,1990;Whitingetal.,1999;Ogrisetal.,2004).InjectioninoocytesormammaliancelllinesofcRNAcodingforα-andβ-subunitsresultsintheexpressionoffunctionalGABAA-RssensitivetoGABA.However,coexpressionofaγ-subunitisrequiredforbenzodiazepinemodulation.IthasrecentlybeensuggestedthatPKCΕregulatesthesensitivityofGABAAα1β2γ2receptorstoethanolandbenzodiazepinesthroughphosphorylationofserine327inthelargeintracellularloopofγ2(Qietal.,2007)
