Theantibodyispreparedfrompooledrabbitserumbyaffinitypurificationusingacolumntowhichthefusionproteinimmunogenwascoupled.

Gamma-aminobutyricacid(GABA)istheprimaryinhibitoryneurotransmitterinthecentralnervoussystem,causingahyperpolarizationofthemembranethroughtheopeningofaCl−channelassociatedwiththeGABAAreceptor(GABAA-R)subtype. GABAA-Rsareimportanttherapeutictargetsforarangeofsedative,anxiolytic,andhypnoticagentsandareimplicatedinseveraldiseasesincludingepilepsy,anxiety,depressionandsubstanceabuse. TheGABAA-Risamultimericsubunitcomplex.Todatesixas,fourbsandfourgs,plusalternativesplicingvariantsofsomeofthesesubunits,havebeenidentified(OlsenandTobin,1990;Whitingetal.,1999;Ogrisetal.,2004). InjectioninoocytesormammaliancelllinesofcRNAcodingfora-andb-subunitsresultsintheexpressionoffunctionalGABAA-RssensitivetoGABA.However,co-expressionofag-subunitisrequiredforbenzodiazepinemodulation. Thevariouseffectsofthebenzodiazepinesinbrainmayalsobemediatedviadifferenta-subunitsofthereceptor(McKernanetal.,2000;MehtaandTicku,1998;Ogrisetal.,2004;Pöltletal.,2003). Morerecentlytherehavebeenanumberofstudiesdemonstratingthattheδ-subunitofthereceptormayaffectsubunitassembly(Korpietal.,2002)andmayalsoconferdifferentialsensitivitytoneurosteroidsandtoethanol(Wallneretal.,2003;Wohlfarthetal.,2002).
