BIOLOGicalSignificance:Collapse
Themetabotropicglutamatereceptors(mGluRs)arekeyreceptorsinthemodulationofexcitatorysynaptictransmissioninthecentralnervoussystem.Theyareimplicatedinmanyformsofneuralplasticityaswellaslearningandmemoryanddrugabuse(Bhattacharyaetal.,2004;Francesconietal.,2004;WilsonandNicoll,2001).GroupImetabotropicglutamatereceptors(consistingofmGluR1andmGluR5)areG-protein-coupledneurotransmitterreceptorsthatarelocalizedintheperisynapticregionofthepostsynapticmembrane.Whenactivated,GroupImGluRsleadtostimulationofphospholipaseandactivationofProteinKinaseC.Incontrast,activationofGroupIImetabotropicreceptors(mGluR2andmGluR3)leadstoinhibitionofadenylatecyclase.ThemGluR1receptormayalsobecriticallyinvolvedinlimitingthedeleteriouseffectsofexcitotoxicity(Blaabjergetal.,2003).
