TheionchannelsactivatedbyglutamatethataresensitivetoN-methyl-Daspartate(NMDA)aredesignatedNMDAReceptors(NMDAR).TheNMDARplaysanessentialroleinmemoryandneuronaldevelopmentandithasalsobeenimplicatedinseveraldisordersofthecentralnervoussystemincludingAlzheimer’s,epilepsyandischemicneuronalcelldeath(Grosshansetal.,2002;Wentholdetal.,2003;CarrollandZukin,2002).ThereareanumberofdifferentsplicevariantsoftheNR1-Subunit(Foldesetal.,1994;ZukinandBennett,1995).DifferentialsplicingofthreeexonsintheNR1-SubunitgeneratesuptoeightNR1-Subunitsplicevariantsand7ofthesehavebeenidentifiedinCDNAlibraries.Theseexonsencodea21aminoacidN-terminaldomain(N1)andadjacentsequencesintheC-terminus(C1andC2).SplicingouttheC2cassetteeliminatesthefirststopcodonandproducesanewreADIngframethatgeneratesanewsequenceof22aminoacids(C2′).ConsiderableattentionhasbeenfocusedonthedistributionandexpressionofthesesplicevariantsthatmayaffectthefunctionalpropertiesandregulationoftheNMDAR
