BIOLOGicalSignificance:Collapse
TheNMDAreceptor(NMDAR)playsanessentialroleinmemory,neuronaldevelopmentandithasalsobeenimplicatedinseveraldisordersofthecentralnervoussystemincludingAlzheimer’s,epilepsyandischemicneuronalcelldeath(Grosshansetal.,2002;Wentholdetal.,2003;CarrollandZukin,2002).TheNMDAreceptorisalsooneoftheprincipalmoleculartargetsforalcoholintheCNS(Lovingeretal.,1989;Alvestadetal.,2003;Snelletal.,1996).TheNMDARisalsopotentiatedbyproteinphosphorylation(Luetal.,1999).TheratNMDAR1(NR1)wasthefirstsubunitoftheNMDARtobecloned.TheNR1proteincanformNMDAactivatedchannelswhenexpressedinXenopusoocytesbutthecurrentsinsuchchannelsaremuchsmallerthanthoseseeninsitu.ChannelswithmorephysiologicalcharacteristicsareproducedwhentheNR1subunitiscombinedwithoneormoreoftheNMDAR2(NR2A-D)subunits.
