TheNMDARplaysanessentialroleinmemory,neuronaldevelopmentandithasalsobeenimplicatedinseveraldisordersofthecentralnervoussystemincludingAlzheimer’s,epilepsyandischemicneuronalcelldeath(Grosshansetal.,2002;Wentholdetal.,2003;CarrollandZukin,2002).TheratNMDAR1(NR1)wasthefirstsubunitoftheNMDARtobecloned.TheNR1proteincanformNMDAactivatedchannelswhenexpressedinXenopusoocytesbutthecurrentsinsuchchannelsaremuchsmallerthanthoseseeninsitu.ChannelswithmorephysiologicalcharacteristicsareproducedwhentheNR1subunitiscombinedwithoneormoreoftheNMDAR2(NR2A-D)subunits(Ishiietal.,1993).PhosphorylationofTyr1336isthoughttopotentiateNMDAreceptor-dependentinfluxofcalcium(Takasuetal.,2002)andischemiamayalsoincreasethephosphorylationofthissite(Takagietal.,2003).
