BIOLOGicalSignificance:Collapse
TheionchannelsactivatedbyglutamatethataresensitivetoN-methyl-D-aspartate(NMDA)aredesignatedNMDAreceptors(NMDAR).TheNMDARplaysanessentialroleinmemory,neuronaldevelopmentandithasalsobeenimplicatedinseveraldisordersofthecentralnervoussystemincludingAlzheimer’s,epilepsyandischemicneuronalcelldeath(Grosshansetal.,2002;Wentholdetal.,2003;CarrollandZukin,2002).TheNMDAreceptorisalsooneoftheprincipalmoleculartargetsforalcoholintheCNS(Lovingeretal.,1989;Alvestadetal.,2003;Snelletal.,1996).TheratNMDAR1(NR1)wasthefirstsubunitoftheNMDARtobeclonedanditcanformNMDAactivatedchannelswhenexpressedinXenopusoocytesbutthecurrentsinsuchchannelsaremuchsmallerthanthoseseeninsitu.ChannelswithmorephysiologicalcharacteristicsareproducedwhentheNR1subunitiscombinedwithoneormoreoftheNMDAR2(NR2A-D)subunits.OverexpressionoftheNR2B-subunitoftheNMDAreceptorhasbeenassociatedwithincreasesinlearningandmemorywhileaged,memoryimpairedanimalshavedeficienciesinNR2Bexpression(Claytonetal.,2002a;Claytonetal.,2002b).TheNMDARisalsopotentiatedbyproteinphosphorylation(Luetal.,1999).
