BIOLOGicalSignificance:Collapse
TheSEROtonintransporter(SERT)recyclesserotoninbytransportingitbacktothepre-synapticcell.Itistheprimarytargetformostanti-depressantdrugsandforstimulantssuchasmethamphetamines.SERTisregulatedbyseveralprocesses,includingacyclicGMPsignalingpathwayinvolvingnitricoxidesynthase,guanylylcyclase,andcGMP-dependentproteinkinase(PKG).cGMP-andPKG-mediatedSERTregulationrequiresphosphorylationatthr276(Ramamoorthyetal.,2007).IthasbeensuggestedthatalthoughPKGisinvolvedinthestimulationofSERTatthr276,itdoesnotdirectlyphosphorylatetheresidue,ratheritinitiatesakinasecascadethatleadstoSERTphosphorylationbyanasyetunidentifiedproteinkinase(Wongetal.,2012).Alsooftherapeuticimportance,mutationatthethr276residuehasbeenshowntodecreasethepotencyofavarietyofanti-depressantdrugs,(ZhangYWandRudnickG,2005).
