TheRaspathwayisacriticalsignaltransductioncascadeinvolvedinregulatingcellularproliferation,differentiation,survival,andoncogenictransformation.MembersoftheRafserine/threoninekinasefamilyarekeyintermediatesinthiscascade,functioningtorelaysignalsfromactivatedRastothedownstreamproteinkinasesMEKandERK(Marshall,1996).PreviousstudieshaveshownthatphosphorylationisrequiredforRaf-1activation(DhillonandKolch,2002;Chongetal.,2003).RecentworkhasdemonstratedthatphosphorylationalsoregulatesthedownregulationofRaf(Doughertyetal.,2005)withtwositesparticipating:Ser301andSer642.
