Theinsulinreceptor(IR)isawell-studiedreceptortyrosinekinasecomposedoftwoαsubunits,responsIBLefortheextracellularinsulinbindingsite,andtwoβsubunits,responsibleforintracellularproteinkinaseactivity(Endemannetal.,1990,Chiuetal.,2010).Thebindingofinsulintotheαsubunitsactivatestheintrinsickinaseactivitylocatedintheβsubunitsandsubsequentlyinitiatesacascadeofphosphorylationeventscausingmajorconformationalchangesintheactivationloopofthekinasedomain,whichleadtodifferentbiologicalfunctions(Chiuetal.,2010).IthasbeenhypothesizedthatT1160phosphorylationaffectsorisaffectedbyY1158/62/63phosphorylationandthattheconformationofT1160andpT1160islikelytobeaffectedbythephosphorylationstatusofthesurroundingtyrosines.
