p70S6kinase(p70S6K)isactivatedinasignalingpathwaythatincludesmTORand isamitogen-activatedSer/ThrproteinkinasethatisrequiredforcellgrowthandG1cellcycleprogression(Xioetal.,2009).p70S6Kiscontrolledbymultiplephosphorylationeventslocatedwithinthecatalytic,linkerandpseudosubstratedomainsandsubsequentlyphosphorylatesspecificallyribosomalproteinS6(Saitohetal.,2002).PhosphorylationofThr229inthecatalyticdomainandThr389inthelinkerdomainaremostcriticalforkinasefunction.Inhibitionofp70activityinhibitstheentryintoSphaseofthecellcycleandexhibitscellcyclearrestatG0/G1phase,suggestingthattheactivationofp70S6kplaysanobligatoryroleinmediatingmitogenicsignalsduringcellactivation(Xioetal.,2009).
