BIOLOGicalSignificance:Collapse
Pantothenatekinase,PanK,isavitalregulatoryenzymeforcoenzymeA(CoA)biosynthesis,phosphorylatingpantothenate(vitaminB5)to4’-phosphopantothenate,thenquicklytransformingtoCoAwhichisanessentialcomponentforfattyacidmetabolism(ABIko,Y,1967). Thereare4membersofthePanKfamily,locatedonchromosomes10q23.31,20p13,5q35,and1p36.32(Zhouetal,2001). PanK1ispredominantlyinheart,liver,andkidney.PanK2isexpressedubiquitously,withhigherlevelsinretinalandinfantbasalganglia. PanK3hashighlevelsinliver,whilePanK4isexpressedubiquitouslywithitshighestlevelsfoundinmuscle(Zhouetal,2001). Additionally,PanK4hasbeenshowntoregulatePkm2activityaffectingglucosemetabolism(Lietal,2005). Therehavebeenseveralphospho-serine,threonine,andtyrosinesitesidentifiedwithinPanK4,theroleofeachonehasyettobedetermined.
