RibosomalproteinS6(rpS6)isacriticalcomponentofthe40Sribosomalsubunitthatmediatestranslationinitiationatthe5′-m7GpppGcapofmRNA.TherpS6proteinisbothcytoplasmicandnuclearlocalized(ChenandDittmer2011).Inresponsetomitogenicstimuli,rpS6undergoesorderedC-terminalphosphorylationbyp70S6kinasesandp90ribosomalS6kinasesonfourSerresidues(Ser235,Ser236,Ser240,andSer244)whosemodificationpotentiatesrpS6capbindingactivity(Hutchinsonetal.,2011).Additionally,rpS6phosphorylationandfunctionarehighlyregulatedandhavebeenimplicatedintheregulationoftranslationalinitiationandproteinsynthesisinresponsetoextracellularstimulisuchasTRAILandgammainterferon(IFN-γ),aswellasuponactivationofthephosphatidylinositol3-kinase(PI3K)–Akt–mTORpathway(ChenandDittmer2011).
