Invitro,TAO(thousandandoneaminoacid)proteinkinase2(TAO2)activatesMAP/ERKkinases(MEKs)3,4,and6towardtheirsubstratesp38MAPkinaseJNK/SAPK(Chenetal.,1999;ChenandCobb,2001).ThisandmorerecentworkhasledtotheproposalthattheTAOproteinkinasesplayanessentialroleinsignalingfromphysiologicalagoNISTstothestress-responsivep38MAPKs(Chenetal.,2003).AutophosphorylationofTAOmayplayaroleinthemechanismofTAOactivation.TheMEKbindingdomainofTAOisautophosphorylatedonbothserineandthreonineresiduesandSer181islocatedwithinthisdomain.
