BIOLOGicalSignificance:Collapse
Synaptojaninisaphosphatidylinositolphosphataseinvolvedinclathrin-mediatedendocytosisofsynapticvesicles.Synaptojanin1hastwoalternativelysplicedisoforms;onethatis~145kDAandisexclusivelyexpressedinneurons,andtheother~170kDawhichisexpressedinnon-neuronal,peripheraltissues(RamjaunAR&McPhersonPS,1996).ThegenewhichencodesSynaptojanin1,SYNJ1,hasbeenmappedtochromosome21thusmakingitacandidateforinvolvementinDown’ssyndrome(DS).IthasrecentlybeendemonstratedthatTs65Dnmice(themostcommonlyusedmodelofDS)havealteredphosphatidylinositol-4,5-bisphosphatemetabolism.ThisdefectisrescuedbyrestoringSYNJ1todisomyintheTs65Dnmice(VoronovSVetal.,2008).
